Masehe escaped a sanction as the referee bizarrely booked Putsche, who had to be replaced by fellow midfielder Thabo Nodada moments later in Bethlehem.
Putsche has since been ruled out of Sunday's MTN8 semi-final decider with Mamelodi Sundowns at Lucas Moripe Stadium and is set to undergo a scan.
The Citizens have asked SAFA's refereeing committee to review the incident.
City said via Twitter: 'Roland is still unable to walk today after the horrific stomp by Paulus Masehe. Putsche is likely to undergo a scan as soon as the team arrives in Johannesburg for the MTN8 semi final second leg. We wish him a speedy recovery!
'The club has made an urgent request to the SAFA refereeing committee to review the incident and the inappropriate sanction that followed.'
Meanwhile, the coastal outfit also confirmed winger Riyaad Norodien is available for the clash with Sundowns after missing the previous two matches with a knock.
Benni McCarthy's side won the first leg 1-0 in the Mother City.
