Macuphu was involved in a car accident on Sunday and is still at home recovering from minor injuries sustained.
The former Chippa United striker is set to miss Matsatsantsa's next game against Sundowns on April 7 due to suspension after he picked up his fourth yellow card against Kaizer Chiefs at Mbombela Stadium last week Friday.
Tembo has termed the absence of Macuphu for their next league match as a "big blow" for his team.
"It’s a big blow but what is disappointing is how he got the yellow card from a long throw-in. Okay it’s a foul, the referee gives a free-kick and then why do you book him?" said Tembo.
"I don’t think it was a bookable offence for me. It's a big blow because he has been scoring goals for us and now we miss him against Sundowns, but we will try to soldier on and see who can replace him," he added.
The Tshwane derby between Sundowns and SuperSport at Loftus Stadium on April 7 remains in doubt due to the former's involvement in the CAF Champions League on the same weekend.
